Function

  • OsVIT1 and OsVIT2 may function primarily in flag leaves to mediate vacuolar sequestration of Fe and Zn, due to highly expression levels of OsVIT1 and OsVIT2 in flag leaves rather than embryos.
  • And they might also mediate allocation of Fe/Zn between flag leaves and the grains[1].

Phenotypic analysis

  • Knockdown of the vacuolar Fe transporter gene OsVIT1 or OsVIT2 may be useful to further increase the Fe concentration of seeds [2].

Expression

  • This gene expresses in flag leaf sheaths with a significantly high level, but it can also be detected in various tissues/organs including embryos at low levels.
  • Different from OsVIT2, OsVIT1 expresses mainly in flag leaf blades, but also occurs ubiquitously in various tissues/organs including embryos with relatively low levels[1].
  • OsVIT2 was quickly induced in rice seedling roots and shoots after the treatment of 4.5 mM FeSO4, but OsVIT1 was not highly induced.
  • Fe starvation caused downregulated OsVIT2 expression in roots and shoots, and no OsVIT1 response until 7 days after treatment in roots.
  • These data suggest that OsVIT1 may represent a housekeeping mechanism for Fe accumulation in vacuoles, while OsVIT2 functions more in response to external Fe levels [1].

Subcellular localization

  • OsVIT1 and OsVIT2 were localized to vacuolar membrane[1] when they combined with enhanced green fluorescent protein and expressed(Figure 1).

Evolution

  • OsVIT2 locates in chromosome 9, and encodes putative protein as the vacuolar iron transporter with 247 amino acids which shares 79.8% amino acid similarity with the VIT in Arabidopsis.
  • There are five conserved transmembrane domains in OsVIT2 by further analyses.
